Líneas de investigación Nuestra área de investigación tiene por objeto abordar dos cuestiones principales en inmunología. La primera se refiere a los mecanismos de control de tráfico de células T efectoras y regulatorias. Las aplicaciones prácticas de esta investigación, aunque en esta etapa la investigación es principalmente de carácter básico, se puede proyectar hacia una inmunoterapias más informadas en las áreas de enfermedades inflamatorias del intestino (enfermedad de Crohn, colitis ulcerosa) y la posibilidad de "guiar" los linfocitos T citotóxicos a los tumores. Nuestro logro importante en esta área fue el hallazgo de que las células dendríticas (DC), la única de célula capaz de iniciar la respuesta inmune, es responsable de "instruir" células T en cuanto a la ubicación del sitio de entrada del agente patógeno (homing). Nuestro segundo, tema básico / aplicado está relacionada con las células dendríticas inmaduras y su uso (IDC) para la producción de células T reguladoras tolerogénicas específicas de antígeno (Treg). Nuestro principal logro en este tema ha sido el diseño y la utilización de nuevos protocolos para la generación de Treg antígeno-específicas en modelos animales capaces de inhibir la proliferación de células T transgénicas efectoras. Una tercera área de investigación está relacionado con el resultado de la respuesta inmune local en el intestino, ya sea hacia una reacción tolerógeno o inflamatoria. Esto está condicionado por un equilibrio entre tres eventos relacionados; la cinética de la generación de Treg vs Th17, la plasticidad y tasa de conversión de Treg a Th17 y de la tasa de expresión de sus receptores de alojamiento específicos de tejido. En base a esto, se puede investigar el efecto que la conversión de un fenotipo Treg supresor en un efector inflamatoria linaje Th17 tiene sobre el resultado de la respuesta inmune asociados al intestino. A third and newer area of basic research we are undertaking deals with our efforts to understand the salmonids immune system. This research stemmed from our recent success in the development of a vaccine that protects salmons against an intracellular bacterial pathogen. Researh interests Our basic research area is aimed at addressing two major questions in immunology. The first one deals with the mechanisms controlling effector and regulatory T cell traffic. The practical applications of this research, although at this stage mostly basic in nature, can be projected towards a more informed immuno-therapy in the areas of bowel inflammatory diseases (Crohn’s, ulcerative colitis) and the possibility of “guiding” effector cytotoxic T cells to tumors. Our major accomplishment in this area was the finding that dendritic cells (DC), the only cell capable of initiating the immune response, is responsible for “instructing” T cells as to the location of the site of entry of the pathogen (homing). Our second, basic/applied subject is related to the use immature dendritic cells (iDCs) for the production of antigen-specific tolerogenic regulatory T cells (Treg). Our main accomplishment in this topic has been the design and utilization of novel protocols for the generation of antigenspecific Treg in animal models capable of inhibiting the proliferation of antigen-specific transgenic effector T cells A third area is related to the outcome of a local immune response towards either a tolerogenic or inflammatory reaction in the gut. This is conditioned by a balance between three related events; the kinetics in the generation of Treg vs. Th17, the plasticity and rate of Treg to Th17 conversion and on the rate of expression of their tissue-specific homing receptors. Based on this, we aim to investigate the effect that the conversion of a suppressive Treg phenotype into an effector inflammatory Th17 lineage has on the outcome of the gut-associated immune responses.
