Hospital Arnau de Vilanova, Lleida, Spain INTRODUCTION Proliferation of vascular smooth muscle cells (VSMC) in parallel with cell senescence takes place in the atherosclerotic process. To be able to divide VSMC need the energy of ATP produced by mitochondria. Alterations in the mitochondrial function are related to increased production of reactive oxygen species (ROS). Among the factors that influence the production of ROS is angiotensin II. Its main role is to control the blood pressure, electrolyte and volume homeostasis, but it has been demonstrated that is also a powerful mediator of oxidative stress, inflammation and cell growth. The presence of a local renin-angiotensin system (RAS) has been shown in the vascular wall. Between the factors that regulates RAS at systemic levels is the active metabolite of vitamin D. The beneficial effects of vitamin D in the cardiovascular system could be due to its ability to decrease the expression of renin. In previous work of our laboratory we found that vitamin D induces an increase in VSMC proliferation and calcification trough a VEGF and RANK-BMP4-dependent pathway, respectively. METHODS AND RESULTS In our current work, we use vitamin D receptor knockout (VDRKO) mice for studies. We found a decrease in the proliferation rates of VSMC in these mice in vitro and in vivo. In quiescent state, Kip2 VDRKO cells presented elevated mRNA expression of p57 together with an augment in the production of angiotensin II in the culture medium in comparison with the wild type (WT). We assessed by RT-PCR and Western blot the expression of cathepsin D, a protease with reninlike activity, and we found a significant increase. By FACS analysis we were able to detect higher production of superoxide anion in the mutant cells, which was inhibited with both losartan and DPI. By high-resolution respirometry we found that the mitochondria of VDRKO VSMC were less efficient, which could explain the lower content of ATP in these cells. Some of the VDRKO cells presented senescence-associated (SA) phenotype at early passage. CONCLUSIONS The lack of signalling trough VDR in VSMC leads to SIPS, probably due to the increased local production of angiotensin II, which point to a possible participation of the vitamin D system in the atherosclerotic process. Laboratorio de Nefrología experimental, IRBLLEIDA, Hospital Arnau de Vilanova, Lleida, Spain INTRODUCCIÓN La proliferación y la senescencia la de las células de músculo liso vascular (CMLV) participan en el proceso aterosclerótico. Para poder dividirse las CMLV necesitan la energía del ATP producido por las mitocondrias. Alteraciones en la función mitocondrial están relacionadas con mayor producción de especies reactivas del oxígeno (ROS). Entre los factores que influyen en la producción de ROS es la angiotensina II. Su función principal es controlar la presión sanguínea, pero se ha demostrado que también es un mediador de crecimiento celular, estrés oxidativo e inflamación. La presencia del sistema de renina-angiotensina (RAS) se ha demostrado en la pared vascular. Entre los factores que regulan el RAS al nivele sistémico es el metabolito activo de la vitamina D. En trabajos anteriores de nuestro laboratorio hemos encontrado que la vitamina D aumenta la proliferación y la calcificación de las CMLV a través de la vía de VEGF y RANK-BMP4, respectivamente. MÉTODOS Y RESULTADOS En nuestro trabajo actual, utilizamos ratones knockout para el receptor de la vitamina D (VDRKO). Se encontró una disminución en las tasas de proliferación de las CMLV de estos ratones in vitro e in vivo. En estado de reposo, las células VDRKO presentaron niveles elevados de p57 KIP2 , junto con un aumento en la producción de angiotensina II en el medio de cultivo, en comparación con el tipo salvaje (WT). Se evaluó por RT-PCR y Western blot la expresión de la catepsina D, una proteasa con actividad parecida a la de renina, y encontramos un aumento significativo. Mediante análisis por FACS, pudimos detectar una mayor producción de anión superóxido en las células mutantes, que fue inhibida tanto con losartán como con DPI. Por respirometría de alta resolución se determinó que las mitocondrias de VDRKO CMLV fueron menos eficientes, lo que podría explicar el menor contenido de ATP en estas células. Algunas de las células VDRKO presentan un fenotipo asociado a la senescencia. CONCLUSIONES La falta de señalización a través de VDR en CMLV lleva a SIPS, probablemente debido al aumento de la producción local de angiotensina II, que apuntan a una posible participación de la vitamina D en el proceso aterosclerótico.
