S tructure of protein Typical protein (lysozyme) β Sheet Other α Helix β Turn Amino acid The steric structure of the protein is formed by interaction of amino acids. Therefore, the steric structure depends on the sequence of amino acids. The structure of the protein is classified into 4 structures as follows: 1. Primary 2. Secondary 3. Tertiary 4. Quaternary 5 Abs 0 200 Quantitative analysis using UV/Vis spectrometer Benzene ring (280 nm) Almost all proteins have a peak at 280 n, therefore, quantitative analysis of protein can be performed with a UV/Vis spectrometer. An alternative quantitative analysis method is colorimetry using reagent (such as the Lowry method , Biuret method, etc.) 250 300 350 400 Wavelength[nm] Concentration [%, M etc.] Quantitative analysis Abs Abs Calibration curve Concentration [%, M etc.] First of all, spectra of each protein concentration are measured to produce a calibration curve. Next, unknown samples are measured and quantitative analysis is performed using the calibration curve. IR spectrum of typical protein Amid I peak contains the secondary structure information (α-Helix, βSheet etc.) Using this peak, the IR-SSE program calculates SSE by PCR ( Principal Component Regression) or PLS (Partial Least Square) method. 1654 0,6 0,4 0,2 0,0 -0,2 -0,4 -0,6 -1,0 1632 -0,8 1686 Principal Component 1, Loadings 0,8 -1,2 1700 1680 1660 1640 1620 cm -1 Loadings plot of Factor 1 (Principal Component 1) for the score plot in the previous Figure. 1600 35 β-Sheet Structure (%) 30 25 20 15 10 5 0 Controls Infected Bar diagram for the means of ß-sheet percentages measured for healthy control and scrapie-affected samples from genetically selected animals.
