Facultad de Ciencias Farmaceúticas, Bioquímica y Biotecnológicas. UCSMArequipa RESUMEN Se aislaron y caracterizaron cuatro diferentes cepas nativas de Aspergillus sp. (A. níger, A. flavus, A. fumigatus y A. spp.) a partir de desechos orgánicos cítricos en diferentes grados de descomposición fungal y mantuvieron un óptimo crecimiento en medio de cultivo agar Sabouraud en períodos de mantenimiento celular de 3 a 5 días termostatizados a una temperatura de 30±0,1°C. Utilizando un CSTR de 1 l. de capacidad en operación discontinua (batch) se determinó la cinética fungal y parametrización de cada una de ellas utilizando para su completa adaptación caldo peptonado a 100 rpm, 1,5 vvm aire estéril y pH 5,5. El modelo peletizado permitió determinar que la cepa A. fumigatus fue la que reportó la mayor cinética de crecimiento sobre sus co-especies determinándose un γ de 0,0017 g.0,3333 .s-1 que resultó ser un 60% mayor que aquellas obtenidas de A. níger y A. flavus (0,00067 g.0,3333 .s-1 ) y un 40% mayor a la obtenida de A. spp (0,0010 g.0,3333 .s-1) definiendo de esta manera a esta cepa como la de mejor comportamiento frente a su aumento de biomasa. Debido a la fuerte demanda de oxígeno de cada una de esta cepas, ya que resultaron ser aerobias estrictas, se determinó su velocidad de transferencia de oxígeno al medio de crecimiento (OTR, Oxygen Transfer Rate) y de consumo por el micelio (OUR, Oxygen Up-take Rate) para establecer la etapa de gobierno de aumento de biomasa que se consideró de interés para el trabajo. Los valores obtenidos de OTR variaron entre 0,30 y 0,40 mg.O2.l-1.h-1 y los de OUR entre 6,00 y 18,00 mg.O2.l-1.h-1 para todas las cepas analizadas con lo que el suministro de aire debió ser continuo y permanente. Estos valores permitieron establecer que la etapa dominante fue la cinética de aumento de biomasa fungal. Determinada esta capacidad de crecimiento, se utilizó un Agua Residual Industrial (ARI) obtenida de una empresa local, para adaptar estas cepas a un contenido elevado de metales pesados. La caracterización de las ARI´s utilizadas, por voltametría, mostró un contenido de Cu(II) de 13,6272 ppm; 0,9833 ppm de Zn(II); 0,0124 ppm de Cd(II) y 0,9114 ppm de Pb(II) demostrando así su alta contaminación por fuentes antropogénicas que resultaron deseables de ser bioremediadas para reducir el impacto ambiental que ocasiona su libre descarga al medioambiente. La configuración y uso de bioreactores con células inmovilizadas continuos resultó ser la mejor alternativa para conformar sistemas de bioremediación de estos metales pesados utilizando las ARI´s obtenidas anteriormente caracterizadas como altamente polutas. El reactor biológico utilizado para este fin se denómino “pulsante” (RBP) en el cual se evaluó su tiempo de retención hidraúlica (TRH; 8,22 h.) y la biodegradación conseguida resultó ser del orden del 97,1% y 8,4 % para Cu y Pb utilizando A.níger; 85,2% y 35,3% utilizando A.flavus; 88,8% y 5,0% utilizando A. fumigatus y del orden del 84,2% y 24,8% para A. spp., respectivamente con lo que se determinó que todas las cepas fungales nativas resultaron afines a estos dos metales pesados reportando ninguna degradación significativa con el Zn y Cd. El diseño final del sistema de ingeniería para la degradacion biológica de ARI’s contaminadas con metales pesados utilizando estas cepas fungales nativas permitió la construcción de una secuencia de cuatro reactores biológicos piloto PFTR (Plug Flow Tubular Reactor) con sistema pulsante (RBP) de 4,80 m. de altura y 4 cm. de diámetro en SS-316L enchaquetado para su termostatización y rellenos con cada una de las cepas fungales aisladas, caracterizadas y adaptadas entrampadas en alginato de calcio (Figura adjunta). La operación continua de este sistema (30°C; 8,22 h. TRH) permitió la degradación de Cu(II) en un 97,21 % y de Pb(II) en un 8,10% de las ARI´s alimentadas confirmando su escasa o nula afinidad por el Cd y Zn. ABSTRACT Four different native strains of Aspergillus sp. (A. niger, A. flavus, A. fumigatus and A. spp.) were isolated and characterized from organic waste derived of citrus with different degrees of decomposition and maintained optimal fungal growth on agar Sabouraud culture medium in maintenance periods of 3 to 5 days thermostated at temperature of 30 ± 0.1 ° C. Using a 1-liter CSTR capacity in discontinuous operation (batch) was determined fungal kinetics and parameterizing each for complete adaptation using peptonized broth at 100 rpm, 1.5 vvm and pH 5.5 sterile air. The pelletized model allowed to determine that the strain A. fumigatus was the one who reported the highest growth kinetics of their cospecies determined a 0.0017 γ G.0, 3333. s-1 which was a 60% higher than those obtained from A. and A. niger flavus (0.00067 G.0, 3333. s-1) and 40% greater than that obtained from A. spp (0.0010 G.0, 3333. s-1) thus defining this strain as the best behavior in the increaseaofbbiomass. Due to the strong demand for oxygen in each of the strains, which proved to be strict aerobes, determined its oxygen transfer rate to the growth medium (OTR, Oxygen Transfer Rate) and consumption by the mycelium (OUR, Oxygen Up-take rate) to set the stage for increased biomass government that was considered of interest for the job. OTR values obtained ranged between 0.30 and 0.40 mg.O2.l-1.h-1 and those between 6.00 and 18.00 OUR mg.O2.l-1.h-1 for all strains tested so that the air supply must be continuous and permanent. These values are allowed to establish that the dominant phase kinetics was increasedafungalnbiomass. Determined this growth capacity, we used an industrial wastewater (ARI) obtained from a local company, to adapt these strains to a high content of heavy metals. The characterization of the ARI's used for voltammetry showed a content of Cu (II) 13.6272 ppm 0.9833 ppm of Zn (II), 0.0124 ppm Cd (II) and 0.9114 ppm Pb (II) demonstrating its high pollution resulting from anthropogenic sources bioremediadas be desirable to reduce the environmental impact caused by their free download to the environment. And biodegradation was achieved on the order of 97.1% and 8, 4% for Cu and Pb A.niger using, 85.2% and 35.3% using A. flavus, 88.8% and 5.0% using A. fumigatus and the order of 84.2% and 24.8% for A. spp. respectively thereby determined that all native fungal strains were similar to these two heavy metals reported no significant degradation in Zn and Cd. The final system design engineering for the biological degradation of ARI's contaminated with heavy metals using these native fungal strains allowed the construction of a sequence of four pilot bioreactors PFTR (Plug Flow Tubular Reactor) with pulsed system (RBP) of 4.80 m. and height 4 cm. of SS-316L diameter jacketed for thermostatting and fills each of the fungal strains isolated, characterized and adapted entrapped in calcium alginate (Figure below). Continued operation of this system (30 ° C; 8.22 h. TRH) allowed the degradation of Cu (II) in a 97.21% and Pb (II) at 8.10% of the ARI's fed confirming their little or no affinity for Cd and Zn.
