No. H Release Date: 07-Aug-2014 ENGLISH Intended Use Antigen retrieval solutions are intended for in vitro diagnostic use to recover antigenicity in formalinfixed, paraffin-embedded tissue. Summary and Explanation The Antigen Retrieval technique† is a novel method for the rescue of antigens from formalin-fixed, paraffin-embedded tissue. It consists of heating sections in a microwave oven in the presence of an antigen retrieval solution. The quality of the staining result is largely dependent on strict adherence to the antigen retrieval protocol. If the antigens are incompletely retrieved, the staining is light and the background may be high. A user following the standard protocol outlined on the reverse side will see crisp, clear nuclear staining with little or no background. Principles of the Procedure Formalin fixation of the tissues induces protein cross links that helps in maintaining the cellular morphology by inactivating the digestive enzymes and preserving the cytoskeleton. At the same time it can cause conformational changes in the epitopes. This may cause a loss of reactivity of a specific antibody to that antigen. High-energy treatment of such proteins in appropriate pH leads to restoration of the epitope structure and hence retrieves the reactivity of antibody to the target antigen. This process is defined as Antigen Retrieval. Shi et al 1991, have suggested that antigen retrieval loosens or breaks the cross linkages induced by formalin. This allows for the enhanced penetration of antibodies and accessibility of epitopes. Reagents Provided One of the following: Cat. Microwave Pressure Cooker ®, Cat. No. 62104, Nordic Ware®, Minneapolis,MN; BioGenex Cat. No. NW001-PC(Optional). • Add 4950 ml deionized water to the carboy. Cap the carboy tightly and shake the carboy for several seconds to completely mix the solution. • Store with cap tightly secured. The working solution may be stored and used at room temperature for up to 1 month. • BioGenex Manual Protocol Standard Protocol 1. Rinse slides in deionized water. Place slides in a plastic staining holder** with any empty slots filled with blank slides. Place the holder in a slide bath containing sufficient volume of the antigen retrieval solution to adequately cover the tissue on the slides. 2. Turn the oven on high power (500-1000 watts) and closely watch the solution until it comes to a rapid boil, and then turn off the oven. (Note: It usually takes 2-4 minutes before a boil is reached. However, the amount of time required may vary significantly depending on a number of factors, such as the starting temperature of the retrieval solution, the wattage of the microwave oven, and the age of the oven (It is very important that a rapid boil is reached for every run before proceeding to the next step.) 3. Set oven power to approximately 200W and simmer for 10 to 15 minutes. (This power setting should be noted and used for this step in all subsequent runs for the same antibody. Each antibody should be tested for the optimal time for this step.) 4. Remove the slide bath from the microwave oven. Allow slides to cool for 20-30 minutes at room temperature. Rinse with several changes of deionized water. Place slides in 1X PBS and continue with the immunostaining procedure. Alternate Method: Microwave Pressure Cooker Protocol The alternate method uses a pressure cooker inside of the microwave. The pressure cooker that we recommend is available through Nordic Ware®, or you may purchase it through BioGenex for your convenience.* 1. Place slides in a slide holder** filling empty slots with blank slides. Put the holder in the slide bath containing 250 ml of working strength Antigen Retrieval Solution. 2. Fill a plastic pressure cooker with 600 ml of distilled water. Up to three baths may be placed side-byside in the pressure cooker. Cover the pressure cooker according to the manufacturer’s instructions. 3. Position the pressure cooker at the center inside the microwave oven, and turn the oven on high power (500-1000 watts) for approximately 15 minutes, until the cooker is fully pressurized. 4. Reduce the microwave power level to 300-350 watts (40% power for 800watt ovens) and allow the solution to simmer for 15 minutes. Then remove the pressure cooker from the microwave oven and carefully open the cover according to the manufacturer’s instructions. Leaving the slides in the cooker, allow the slides to cool for 20 minutes, then continue with the immunostaining procedure. Quality Control A positive control slide, which has been treated with the same antigen retrieval conditions as the tissues of interest, should be included in every experiment. This product has been validated through extensive testing on the following tissues: stomach, kidney, liver and skin. Run appropriate controls slides with and without antigen retrieval. The antigen retrieval should enhance the signal with appropriate control antibody. Trouble shooting 1. Avoid allowing the solution to boil over as this may cause the tissue to come off the slide or the tissue to dry out. 2. Some tissues are more prone to coming off the slides during antigen retrieval (particularly fatty tissue). Should this occur it might be necessary to bring the antigen retrieval solution to a boil prior to adding the slides, followed by a gentle simmer for 3-5 minutes. Then cool to room temperature for 20-30 minutes prior to continuing with immunohistochemistry. 3. Non-specific staining in the negative control can be due to the exposure of endogenous biotin. Over retrieval can sometimes make the tissue act like frozen tissue. This will make the detection system too strong or the antibody too concentrated. Further titration may be necessary. Including an additional avidin-biotin block should prevent staining of the exposed biotin. 4. After antigen retrieval, the slides can be stored overnight in Phosphate Buffered Saline(PBS)at 4ºC if required. This can, however, cause some tissues to fall off and care should be taken to avoid this. 5. If positive control gives optimum signal, negative control shows no background and test slide gives negative or weak signal, a different fixative might have been used. In order to obtain the best signal in tissue sections fixed with a different fixative, optimization of antigen retrieval conditions is recommended. 6. Refer to appropriate antibody and detection system inserts for pattern and intensity of staining with different antibodies. Expected Results Antigen retrieval can produce markedly improved staining of a wide variety of monoclonal and polyclonal antibodies. This helps overcome false negative staining of over fixed tissue, expand the range of antibodies that can be used and increase the usefulness of archival tissue. Storage and Handling Store all reagents at 2-8°C. Do not use after expiration dates as indicated on the reagent labels. Optimized antigen retrieval should improve signal to noise in immunohistochemistry. Specimen Preparation Appropriate fixation plays an important role in preserving the tissue structure. The antigen retrieval protocol is recommended for use in tissues that have been fixed in formalin only. Ensure that the fixed sections are adequately embedded in paraffin. Cut tissue sections to 4-5 microns. Limitations of the procedure The antigen retrieval protocol is recommended for use with tissues fixed with formalin only. Other fixatives or fixation procedures may not produce comparable results. Interpretation of the staining results is solely the responsibility of the user. Preparation of Working Solutions Antigen Retrieval Solution, are available in a ready to use (RTU) or a concentrated format. The 10X concentrated format should be diluted tenfold with deionized water (mix one part of concentrated Antigen Retrieval Solution with nine parts of deionized water). EZ-DeWax™ Solution is concentrated and requires reconstitution in 500ml reagent alcohol. • For AR1, AR2 and AR3, prepare as stated below: • Shake the bottle vigorously to completely mix the components of the concentrate (the solution may separate into phases over time). • Pour the entire contents of the bottle into the corresponding AR carboy. Add 225 ml histologicalgrade alcohol to the carboy. (Note: AR solutions must be reconstituted with histological-grade alcohol. For example, Fisher Reagent Grade Alcohol, Cat. No. A962, or Sigma Reagent Alcohol, Cat. No. R8382, are specifically denatured for histological use and also specifically filtered for use in tissue processors.) Assicurarsi che le sezioni fissate siano opportunamente incluse in paraffina. Tagliare sezioni di tessuto di 4-5 micron. Per esempio, l’alcool della Fisher, codice A962, o quello della Sigma, codice R8382, sono specificamente denaturati per uso istologico ed anche specificamente filtrati per l’uso in processatori di tessuto). • Aggiungere 4.950 ml di acqua deionizzata alla tanica. Chiudere bene la tanica con il relativo tappo e scuotere la tanica per alcuni secondi per completare il mescolamento della soluzione. • Conservare con il tappo ben chiuso. La soluzione di lavoro può essere conservata ed usata a temperatura ambiente fino ad 1 mese dalla sua ricostituzione. Protocollo manuale BioGenex Protocollo standard 1. Risciacquare i vetrini in acqua deionizzata. Collocare i vetrini in un supporto di plastica per colorazione**, in cui gli eventuali slot vuoti siano riempiti con vetrini inutilizzati. Collocare il supporto in una vaschetta per vetrini, contenente soluzione per antigen retrieval in quantità sufficiente da coprire opportunamente il tessuto sui vetrini. 2. Portare il forno ad alta potenza (500-1000 watt) e osservare da vicino la soluzione finché non giunge a rapida ebollizione, quindi spegnere il forno. (Nota: occorrono in genere 2-4 minuti prima che si raggiunga l'ebollizione. Tuttavia, la quantità di tempo necessaria può variare sensibilmente in funzione di numerosi fattori, come la temperatura iniziale della soluzione di recupero, il wattaggio del forno a microonde e l'età del forno (è molto importante che si raggiunga un'ebollizione rapida per ogni ciclo, prima di procedere alla fase successiva). 3. Regolare la potenza del forno su circa 200 watt e scaldare per 10-15 minuti. (È bene annotare questo valore di potenza e utilizzarlo per questa fase, in tutti i cicli successivi per lo stesso anticorpo. Occorre verificare per ogni anticorpo quale sia la quantità di tempo ottimale per questa fase). 4. Rimuovere la vaschetta con i vetrini dal forno a microonde. Lasciar raffreddare i vetrini per 20-30 minuti a temperatura ambiente. Risciacquare più volte i vetrini con acqua deionizzata. Collocare i vetrini in soluzione tampone fosfato (PBS) 1X e continuare con la procedura di immunocolorazione. Metodo alternativo: protocollo Microwave Pressure Cooker Il metodo alternativo si avvale di una pentola a pressione invece del forno a microonde. La pentola a pressione da noi consigliata è disponibile presso la Nordic Ware® o, eventualmente, può essere acquistata tramite la BioGenex. 1. Collocare i vetrini in un supporto per colorazione**, riempiendo gli slot vuoti con vetrini inutilizzati. Collocare il supporto in una vaschetta per vetrini, contenente 250 ml di soluzione di lavoro per antigen retrieval. 2. Riempire una pentola a pressione di plastica con 600 ml di acqua distillata. È possibile inserire nella pentola a pressione fino a tre vaschette, una di fianco all'altra. Chiudere la pentola a pressione secondo le istruzioni fornite dal produttore. 3. Posizionare la pentola a pressione al centro del forno a microonde, portare il forno ad alta potenza (500-1000 watt) per circa 15 minuti, finché la pentola non sia interamente sotto pressione. 4. Ridurre la potenza del forno a 300-350 watt (40% della potenza per i forni da 800 watt) e lasciar bollire lentamente la soluzione per 15 minuti. Quindi togliere la pentola a pressione dal forno e rimuovere con cautela il coperchio, secondo le istruzioni fornite dal produttore. Senza togliere i vetrini dalla pentola, lasciarli raffreddare per 20 minuti, quindi continuare con la procedura di immunocolorazione. Controllo qualità In ogni esperimento deve essere incluso un vetrino di controllo positivo, che sia stato trattato con le stesse condizioni di recupero antigenicità dei tessuti d'interesse. Questo prodotto è stato convalidato mediante test approfonditi, condotti sui seguenti tessuti: stomaco, reni, fegato e cute. Utilizzare opportuni vetrini di controllo con e senza antigen retrieval. L'antigen retrieval dovrebbe amplificare il segnale con un idoneo anticorpo di controllo. Risoluzione dei problemi 1. Non lasciar bollire la soluzione per un periodo eccessivo, poiché ciò potrebbe provocare il distacco del tessuto dal vetrino o l'essiccazione del tessuto. 2. Alcuni tessuti sono soggetti a staccarsi dai vetrini durante l'antigen retrieval (soprattutto i tessuti grassi). Qualora ciò accadesse, potrebbe essere necessario portare la soluzione per antigen retrieval ad ebollizione prima di aggiungere i vetrini, lasciandola poi bollire lentamente per 3-5 minuti. Quindi lasciar raffreddare per 20-30 minuti a temperatura ambiente, prima di continuare con l'immunoistochimica. 3. Una colorazione aspecifica nel controllo negativo può essere dovuta all'esposizione della biotina endogena. Talvolta, un recupero eccessivo dell'antigenicità può indurre il tessuto a comportarsi come tessuto congelato. Ciò rende il sistema di rivelazione troppo potente o l'anticorpo troppo concentrato. Potrebbe occorrere un'ulteriore titolazione. Aggiungendo un complesso avidina-biotina, si dovrebbe impedire la colorazione della biotina esposta. 4. Dopo l'antigen retrieval, i vetrini possono essere conservati per la notte, se necessario, in PBS a 4°C. Tuttavia, ciò può causare il distacco di taluni tessuti: adottare ogni precauzione per evitarlo. 5. Se il controllo positivo fornisce un segnale ottimo, il controllo negativo non mostra alcun fondo e il vetrino di prova fornisce un segnale negativo o debole, significa che probabilmente è stato utilizzato un fissativo diverso. Per ottenere il segnale migliore, in sezioni di tessuto fissate con un fissativo diverso, si raccomanda di ottimizzare le condizioni dell'antigen retrieval. 6. Consultare gli appositi foglietti illustrativi degli anticorpi e dei sistemi di rivelazione per conoscere il pattern e l'intensità della colorazione con anticorpi diversi. Risultati attesi L'antigen retrieval può fornire una colorazione notevolmente potenziata di un'ampia varietà di anticorpi monoclonali e policlonali. Si può così ovviare agli inconvenienti della colorazione falsa negativa di un tessuto sottoposto ad eccessiva fissazione, ampliare la gamma degli anticorpi utilizzabili ed accrescere l'utilità del tessuto d'archiviazione. Una tecnica ottimizzata di antigen retrieval dovrebbe migliorare il rapporto segnale/rumore in immunoistochimica. Limiti della procedura Il protocollo di antigen retrieval è raccomandato per l'uso su tessuti che sono stati fissati esclusivamente in formalina. Altri fissativi o altre procedure di fissazione potrebbero non fornire risultati altrettanto validi. L'interpretazione dei risultati della colorazione è di esclusiva responsabilità dell'utilizzatore. Caratteristiche funzionali BioGenex ha condotto diversi studi per valutare l'efficacia di tutti i suoi agenti per antigen retrieval, utilizzando diversi sistemi di rivelazione e anticorpi BioGenex. DEUTSCH, GERMAN Verwendungszweck Antigen Retrieval Lösungen dienen in der In-vitro-Diagnostik zur Wiederherstellung von Antigenität in Formalin-fixiertem, Paraffin-eingebettetem Gewebe. Zusammenfassung und Erklärung Die Antigen-Wiederherstellungstechnik† ist ein neuartiges Verfahren zur Befreiung von Antigenen aus Formalin-fixiertem, Paraffin-eingebettetem Gewebe. Dabei werden Schnitte in einem Mikrowellenherd in Anwesenheit einer Antigen Retrieval Lösung erhitzt. Die Qualität des Färberesultats hängt zum Großteil von der strikten Befolgung des Antigen-Wiederherstellungsprotokolls ab. Wenn die Antigene unvollständig wiederhergestellt werden, ist die Anfärbung hell und der Hintergrund möglicherweise hochrot. Ein Anwender, der das auf der Rückseite beschriebene Standardprotokoll befolgt, sieht eine klare, deutliche Zellkernfärbung mit wenig oder keinem Hintergrund. Prinzipien des Verfahrens Die Formalinfixierung der Gewebe induziert Protein-Quervernetzungen, die helfen, die Zellmorphologie durch Inaktivierung der Verdauungsenzyme und Erhalt des Zytoskeletts aufrecht zu erhalten. Gleichzeitig kann sie Konformationsänderungen in den Epitopen verursachen. Das kann zu einem Reaktivitätsverlust eines spezifischen Antikörpers gegen dieses Antigen führen. Eine Hochenergiebehandlung solcher Proteine bei einem geeigneten pH-Wert führt zur Wiederherstellung der Epitopstruktur und damit auch der Reaktivität des Antikörpers gegen das Zielantigen. Dieser Vorgang ist als „Antigen Retrieval“ (Antigen-Wiederherstellung) definiert. Shi et al., 1991, haben vorgeschlagen, dass die Antigen-Wiederherstellung die durch Formalin induzierten Quervernetzungen lockert oder aufbricht. Zubereitung von Arbeitslösungen Die “Antigen Retrieval” Lösung ist in einem ready to use oder in einem konzentriertem Format verfügbar. Das 10-fach konzentrierte Format muss mit de-ionisiertem Wasser (ein Teil der konzentrierten Antigen Retrieval Lösung wird mit neun Teilen de-ionisiertem Wasser gemischt) 10-fach verdünnt werden. Die EZ-DeWax™ Lösung ist konzentriert und benötigt eine Rekonstituierung in 500 ml Reagenzien-Alkohol. 2. Einen Kunststoff-Druckkochtopf mit 600 ml destilliertem Wasser füllen. Bis zu drei Bäder können Seite an Seite in den Druckkochtopf gestellt werden. Den Druckkochtopf entsprechend den Herstelleranweisungen abdecken. 3. Den Druckkochtopf in die Mitte des Mikrowellenherds stellen und diesen für etwa 15 Minuten auf hohe Leistung (500–1000 Watt) einstellen, bis der Kochtopf vollständig unter Druck steht. 4. Die Mikrowellenleistung auf 300–350 Watt reduzieren (40 % Leistung für 800-Watt-Herde) und die Lösung 15 Minuten köcheln lassen. Den Druckkochtopf dann aus dem Mikrowellenherd nehmen und die Abdeckung vorsichtig entsprechend den Herstelleranweisungen öffnen. Die Objektträger im Kochtopf 20 Minuten abkühlen lassen und dann mit der Immunfärbung weitermachen. Qualitätskontrolle Ein positiver Kontroll-Objektträger, der mit den gleichen Antigen-Wiederherstellungsbedingungen wie die Zielgewebe behandelt wurde, sollte bei jedem Experiment mitlaufen. Dieses Produkt wurde durch ausgedehnte Tests an den folgenden Geweben überprüft: Magen, Niere, Leber und Haut. Geeignete Kontroll-Objektträger mit und ohne Antigen-Wiederherstellung mitlaufen lassen. Die AntigenWiederherstellung sollte das Signal mit geeignetem Kontroll-Antikörper verstärken. Fehlerbehebung 1. Ein Überkochen der Lösung vermeiden, da dies dazu führen kann, dass sich das Gewebe vom Objektträger ablöst oder austrocknet. 2. Einige Gewebe neigen eher dazu, sich während der Antigen-Wiederherstellung von den Objektträgern abzulösen (insbesondere Fettgewebe). Falls dies auftritt, kann es erforderlich sein, die Antigen Retrieval Lösung vor Zugabe der Objektträger zum Kochen zu bringen und dann 3–5 Minuten leicht köcheln zu lasen. Dann 20–30 Minuten auf Zimmertemperatur abkühlen, bevor mit der Immunohistochemie weitergemacht wird. 3. Eine unspezifische Färbung in der Negativkontrolle kann durch Exposition von endogenem Biotin verursacht werden. Eine übermäßige Wiederherstellung kann gelegentlich dazu führen, dass das Gewebe wie gefrorenes Gewebe reagiert. Das macht das Nachweissystem zu stark oder den Antikörper zu konzentriert. Eine weitere Titration kann erforderlich sein. Das Einbeziehen eines zusätzlichen AvidinBiotin-Blocks sollte eine Anfärbung des exponierten Biotins verhindern. 4. Nach der Antigen-Wiederherstellung können die Objektträger falls erforderlich über Nacht in PBS bei 4 °C gelagert werden. Das kann jedoch bei einigen Geweben zur Ablösung führen und sollte möglichst vermieden werden. 5. Wenn die positive Kontrolle ein optimales Signal gibt, die Negativkontrolle keinen Hintergrund zeigt und der Test-Objektträger ein negatives oder schwaches Signal gibt, wurde möglicherweise ein anderes Fixiermittel verwendet. Um bei Gewebeschnitten, die mit einem anderen Mittel fixiert wurden, das bestmögliche Signal zu erhalten, wird eine Optimierung der Antigen-Wiederherstellungsbedingungen empfohlen. 6. Die Informationen über Färbemuster und -intensität bei anderen Antikörpern sind den entsprechenden Packungsbeilagen von Antikörpern und Nachweissystemen zu entnehmen. Erwartete Ergebnisse Die Antigen-Wiederherstellung kann eine deutlich verbesserte Anfärbung einer breiten Palette von monoklonalen und polyklonalen Antikörpern bewirken. Dies hilft, eine falsch negative Anfärbung von überfixiertem Gewebe auszugleichen, die Palette der verwendbaren Antikörper zu vergrößern und die Brauchbarkeit archivierter Gewebe zu steigern. Die optimierte Antigen-Wiederherstellung sollte das Signal/Rausch-Verhältnis in der Immunohistochemie verbessern. Einschränkungen des Verfahrens Das Antigen-Wiederherstellungsprotokoll wird nur für Formalin-fixierte Gewebe empfohlen. Andere Fixative oder Fixierverfahren können möglicherweise nicht vergleichbare Resultate ergeben. Die Interpretation der Färbungsresultate obliegt der alleinigen Verantwortung des Anwenders. • AR1, AR2 und AR3 werden wie folgt vorbereitet: • Schütteln Sie die Flasche heftig um die Komponenten des Konzentrates vollständig durchzumischen (die Lösung kann sich über die Zeit in die einzelnen Phasen separieren). • Füllen Sie den gesamten Inhalt der Flasche in den entsprechende AR-Ballon. • Fügen Sie 225 ml Alkohol histologischen Gütegrades in den Ballon bei (Anmerkung: AR Lösungen müssen in Alkohol histologischen Gütegrades rekonstituiert werden. Zum Beispiel: • Fisher Alkohol Reagenzien-Gütegrad, Cat. No. A962, oder Sigma Alkohol Reagenzien-Gütegrad sind speziell denaturiert für den histologischen Gebrauch und ebenfalls spezifisch für den Einsatz in Gewebe-Verarbeitungssystemen gefiltert). • Fügen Sie 4950 ml de-ionisiertes Wasser in den Ballon bei. • Schliessen Sie den Ballon fest zu und schütteln Sie ihn mehrerer Sekunden lang, um die Lösung vollständig zu mischen. • Lagern Sie die Lösung mit fest geschlossenem Deckel. Objektträger in deionisiertem Wasser abspülen. Objektträger in einen Kunststoff-Färbehalter** stellen, wobei alle leeren Einschübe mit blanken Objektträgern gefüllt werden. Den Halter in ein Objektträgerbad mit einer ausreichenden Menge der Antigen Retrieval Lösung zur Bedeckung des Gewebes auf den Objektträgern stellen. 2. Den Herd auf hohe Leistung (500–1000 Watt) einstellen und die Lösung beobachten, bis es zu einem schnellen Aufkochen kommt, dann den Herd ausschalten. (Hinweis: Es dauert normalerweise 2–4 Minuten bis zum Aufkochen.) Die benötigte Zeit kann jedoch beträchtlich variieren, abhängig von mehreren Faktoren wie beispielsweise der Anfangstemperatur der Wiederherstellungslösung, der Wattzahl des Mikrowellenherds und dem Alter des Herds (es ist sehr wichtig, dass für jeden Durchlauf ein schnelles Aufkochen erreicht wird, bevor mit dem nächsten Schritt fortgefahren wird) 3. Die Herdleistung auf etwa 200 W einstellen und 10 bis 15 Minuten auf köcheln lassen. Jeder Antikörper sollte auf die optimale Zeit für diesen Schritt getestet werden.) 4. Das Objektträgerbad aus dem Mikrowellenherd nehmen. Die Objektträger 20–30 Minuten auf Zimmertemperatur abkühlen lassen. Mit mehrfach gewechseltem deionisiertem Wasser abspülen. Die Objektträger in 1X PBS geben und mit dem Immunfärbeverfahren weitermachen. ESPANOL, SPANISH Resumen y explicación La técnica de Recuperación del antígeno† es un nuevo método para el rescate de antígeno en un tejido fijado con formol y embebido en parafina. Consiste en calentar los cortes en un horno microondas en presencia de una solución de recuperación del antígeno. La calidad del resultado de la tinción depende en gran medida de que se siga estrictamente el protocolo de recuperación del antígeno. Si la recuperación de los antígenos es incompleta, la tinción es clara y puede ser alta en el fondo. El usuario que siga el protocolo estándar que se expone en el reverso de esta hoja verá una tinción nítida y transparente del núcleo con un fondo escaso o nulo. Principios del procedimiento La fijación de los tejidos con formol induce una reticulación proteica que ayuda a mantener la morfología celular al inactivar las enzimas digestivas y conservar el citoesqueleto. Al mismo tiempo, puede provocar cambios de conformación en los epitopos. Con ello, se puede perder la reactividad ante un anticuerpo específico de ese antígeno. El tratamiento de alta energía de tales proteínas en un pH adecuado restaura la estructura de los epitopos y, en consecuencia, se recupera la reactividad del anticuerpo hacia el antígeno diana. Este proceso se define como “recuperación del antígeno”. Shi y cols. sugirieron en 1991 que la recuperación del antígeno debilita o rompe la reticulación inducida por formol. Preparación de las soluciones de trabajo La Solución de Recuperación de Antígenos (Antigen Retrieval Solution) está disponible en dos formatos: lista para usar o concentrada. El formato con una concentración de 10X tiene que diluirse diez veces con agua desionizada (mezcle una parte de Solución concentrada de Recuperación de Antígenos con nueve partes de agua desionizada). La solución EZ-DeWax(tm) es concentrada y tiene que reconstituirse en 500ml de alcohol reactivo. • Para AR1, AR2 y AR3, realice los siguientes pasos: • Agite bien la botella para mezclar correctamente los componentes del concentrado (es posible que, con el tiempo, la solución se haya separado en sus diferentes fases). • Vierta todo el contenido de la botella en el correspondiente bidón de AR. • Añada 225 ml de alcohol para uso histológico al bidón. Tape bien el bidón y agítelo durante algunos segundos, hasta que la solución esté completamente mezclada. • Antes de almacenar el bidón, asegúrese de que está bien tapado. La solución de trabajo puede almacenarse y utilizarse a temperatura ambiente, y como máximo en el plazo de 1 mes. Protocolo manual de BioGenex Protocolo estándar 1. Aclare los portaobjetos con agua desionizada. Coloque los portaobjetos en un soporte de tinción de plástico**, llenando las ranuras vacías con portaobjetos en blanco. Coloque el soporte en un baño de portaobjetos que contenga el volumen suficiente de solución de recuperación del antígeno para cubrir adecuadamente los tejidos en los portaobjetos. 2. Encienda el horno (500-1000 vatios) y vigile de cerca la solución hasta que comience un rápido hervor, y después apague el horno. (Nota: Habitualmente se tarda 2-4 minutos para alcanzar el hervor. No obstante, la cantidad de tiempo requerida puede variar significativamente dependiendo de varios factores, como la temperatura inicial de la solución de recuperación, el voltaje del horno microondas y la antigüedad del horno (es muy importante que se alcance el hervor rápido en cada serie antes de continuar con el paso siguiente). 3. Fije la potencia del horno aproximadamente en 200 W y déjelo hervir durante 10-15 minutos a fuego lento. (Esta configuración se anotará y se usará en este paso en todas las series sucesivas para el mismo anticuerpo. En cada anticuerpo se deberá comprobar el tiempo óptimo de este paso). 4. Extraiga el baño de portaobjetos del horno microondas. Deje enfriar los portaobjetos durante 20-30 minutos a temperatura ambiente. Aclárelos con varios cambios de agua desionizada. Coloque los portaobjetos en PBS 1X y continúe con el procedimiento de inmunotinción. Resolución de problemas 1. No deje hervir la solución en exceso, ya que el tejido podría desprenderse del portaobjetos o secarse. 2. Algunos tejidos son más propensos a desprenderse del portaobjetos durante la recuperación del antígeno (en particular, el tejido graso). Si esto ocurre, podría ser necesario llevar a ebullición la solución de recuperación del antígeno antes de añadir los portaobjetos, dejándolos hervir después suavemente durante 3-5 minutos. Seguidamente, enfríelos a temperatura ambiente durante 20-30 minutos antes de continuar con la inmunohistoquímica. 3. La tinción inespecífica del control negativo puede deberse a la exposición de la biotina endógena. Un exceso de recuperación puede hacer que el tejido actúe como un tejido congelado. Con ello se consigue que el sistema de detección sea demasiado fuerte o que el anticuerpo esté demasiado concentrado. Puede ser necesaria una nueva valoración. La inclusión de un bloqueo adicional de avidina-biotina evitará la tinción de la biotina expuesta. 4. Después de recuperar el antígeno, los portaobjetos se pueden almacenar toda la noche en PBS a 4 ºC, si es necesario. Sin embargo, con ello se puede conseguir que se desprendan algunos tejidos, por lo que se evitará hacerlo. 5. Se deberá usar un fijador diferente si el control positivo da una señal óptima, el control negativo no muestra tinción en el fondo, y el portaobjetos de prueba da una señal negativa o débil. Se recomienda optimizar las condiciones de recuperación del antígeno para obtener la mejor señal en los cortes de tejido fijados con un fijador diferente. 6. Consulte en los prospectos correspondientes del anticuerpo y del sistema de detección el patrón y la intensidad con distintos anticuerpos. Resultados esperados La recuperación del antígeno puede producir una importante mejora de la tinción con varios anticuerpos monoclonales y policlonales. Con ello se evita la tinción negativa falsa del tejido fijado en exceso, y aumenta el número de anticuerpos que se pueden usar y la utilidad del tejido archivado. La recuperación optimizada del antígeno debe mejorar la relación señal-ruido en inmunohistoquímica. Limitaciones del procedimiento Se recomienda usar el protocolo de recuperación del antígeno únicamente con tejidos fijados con formol. Los resultados obtenidos con otros fijadores o procedimientos de fijación pueden no ser comparables. La interpretación del resultado de la tinción es únicamente responsabilidad del usuario. Características de funcionamiento BioGenex ha realizado estudios para evaluar el funcionamiento de todos los agentes de recuperación del antígeno usando varios anticuerpos y sistemas de detección de BioGenex. Ponga el soporte en el baño de portaobjetos con 250 ml de Antigen Retrieval Solution a la concentración de trabajo. 2. Llene la olla a presión de plástico con 600 ml de agua destilada. Se pueden colocar hasta tres baños lado a lado en la olla a presión. Cubra la olla a presión siguiendo las instrucciones del fabricante. 3. Ponga la olla a presión en el centro del horno microondas y enciéndalo a una potencia alta (500-1000 vatios) durante aproximadamente 15 minutos, hasta que se alcance la máxima presión en la olla. 4. Reduzca la potencia del microondas a 300-350 vatios (40% de la potencia para los hornos de 800 vatios) y deje que la solución hierva a fuego lento durante 15 minutos. Extraiga a continuación la olla a presión del microondas y abra la tapa despacio siguiendo las instrucciones del fabricante. Deje enfriar los portaobjetos durante 20 minutos dentro de la olla y después continúe con el procedimiento de inmunotinción. Control de calidad En cada experimento se debe incluir un portaobjetos de control positivo que se haya tratado con las mismas condiciones de recuperación del antígeno que los tejidos de interés. Este producto se ha validado mediante un estudio exhaustivo en los siguientes tejidos: estómago, riñón, hígado y piel. Procese los portaobjetos de control adecuados, con y sin la recuperación del antígeno. La recuperación del antígeno debe mejorar la señal con un anticuerpo de control apropiado. 2
