INTENDED USE:
For the preservation, transportation and the processing of Respiratory specimens such as Sputum and Bronchial specimens to
produce a readable, diagnostic slide to be read and diagnosed by a laboratory professional.
SUMMARY AND EXPLANATION:
The following are the components of this processing Kit:
 Liqui-PREPTM Preservative R Collection Vial - 15mL per vial for the preservation, transport and fixation of the respiratory
specimen.
 Liqui-PREPTM Cleaning Solution R - 245mL per bottle for removal of mucus by density centrifugation from respiratory
specimens.
 Liqui-PREPTM Cellular Base Solution R - 26mL per bottle for encapsulation and adhering of Liqui-PREPTM processed cells onto
a clean glass microscope slide.
STABILITY:
 Supplied reagents and solutions are stable until the labeled expiration date.
 Specimens collected in the Liqui-PREPTM Preservative R Solution is stable for 30 days at 10O to 30O C.

 Swinging bucket style centrifuge (up to 1,200 xg)
 Vortex mixer - minimum of 1,000 RPM
PRINCIPALS OF THE PROCESS:
COLLECTION - up to 10mL of the freshly collected respiratory specimen is placed into the Liqui-PREPTM Preservative R
Collection Vial. The specimen is labeled and sent to the laboratory for processing and diagnosis.
PRESERVATION - Specimens need to reside in the Liqui-PREPTM Preservative R Solution at least 1 hour prior to processing.
CLEANING AND CONCENTRATION - After preservation, the specimen is poured onto 5mL of Liqui-PREPTM Cleaning Solution R,
in a 30mL centrifuge tube and centrifuged for 10 minutes at 1,000 xg Force. The supernatant is discarded resulting in a reasonably
clean, dry cellular pellet.
ENCAPSULATION AND ADHERENCE - The concentrated cell pellet is mixed (vortexed) well to produce a homogeneous
suspension. Take 100µl of this completely homogeneous suspension and place it into a clean dry tube (dilution tube), 500µl of
Liqui-PREPTM Cellular Base Solution is added to the 100µl of homogeneous cell suspension thus producing an appropriate cell
concentration. This final suspension is then mixed very well, using a vortex, immediately prior to taking 50µl of the final
suspension and applying it to a clean dry glass microscope slide. After drying, the Liqui-PREPTM processed slide is ready for
staining and interpretation by a trained professional.
WARNINGS:
 The reagents in this kit contain very dilute mixtures of denatured alcohols and are not intended for human consumption.
NOTES:
 Airborne Pathogen Safety Procedures should be followed when processing any respiratory specimens.
 Specimen concentration or taking random aliquots may be required for specimens of large volume.
 Any specimens not collected in the Liqui-PREPTM Preservative Solution R or Liqui-PREPTM Preservative Solution to be
stored prior to processing should be placed in standard Liqui-PREPTM Preservative Solution (1:2 dilution), if greater than 12
hours store at 2o-8o C.

PRINCIPIOS DEL PROCESO:
COLECCION - hasta 10 ml de la muestra respiratoria recién extraída se coloca en el Liqui-PREPTM Preservative R Collection Vial. La
muestra se rotula y se envía al laboratorio para su procesamiento y diagnóstico.
PRESERVACION - Las muestras deben residir en el Liqui-PREPTM Preservative R Solution al menos 1 hora antes de su procesamiento.
LIMPIEZA Y CONCENTRACION - Después de la preservación, la muestra se vierte en 5 ml de Liqui-PREPTM Cleaning Solution R, en un
tubo de centrífuga de 30 ml y se centrifuga durante 10 minutos a una fuerza 1,000 xg. El sobrenadante se descarta resultando en un botón
celular razonablemente limpio y seco.
ENCAPSULACION Y ADHESION – El botón celular concentrado es mezclado bien (vórtex) para producir una suspensión homogénea.
Tomar 100μl de esta suspensión completamente homogénea y colocar en un tubo limpio y seco (tubo de dilución), se añaden 500μl de
Liqui-PREPTM Cellular Base Solution R a los 100μl de suspensión celular homogénea produciendo así una concentración celular
apropiada. Esta suspensión final se mezcla entonces muy bien, utilizando un vórtex, inmediatamente antes de tomar 50μl de la suspensión
final y aplicándolos a una diapositiva de microscopio de vidrio limpia y seca. Después del secado, la diapositiva procesada de Liqui-PREPTM
está lista para la teñir y para interpretación por un profesional capacitado.
ADVERTENCIAS:

Los reactivos de este kit contienen mezclas muy diluidas de alcoholes desnaturalizados y no están destinados para el consumo humano.
NOTAS:



Procedimientos de Seguridad en el aire los agentes patógenos se deben seguir en el tratamiento de las muestras
respiratorias.
Concentración de muestra o tomar alícuotas al azar puede ser requeridos para especímenes de gran volumen.
Las muestras no recolectadas en Liqui-PREPTM Preservative R Solution o Liqui-PREPTM Preservative Solution
para ser almacenadas antes de ser procesadas deben ser colocadas en la solución estándar de Liqui-PREPTM Preservative Solution
(dilución 1:2), si es mayor de 12 horas almacene a 2°-8° C.

STEP 1: COLLECTION




The specimen from the patient must be transferred immediately into the Liqui-PREPTM Preservative R Collection Vial.
A MAXIMUM OF 10mL OF PATIENT SPECIMEN CAN BE TRANSFERRED INTO THE Liqui-PREPTM Preservative R Collection Vial.
After collection, label the collected specimen and send it to the laboratory for processing and diagnosis.
Excess patient specimen should be stored by adding 1 part of specimen to 2 parts of Liqui-PREPTM Preservative Solution at 2O-8O C.
PASO 1: COLECCION




La muestra del paciente debe ser transferida inmediatamente en Liqui-PREPTM Preservative R Collection Vial.
UN MAXIMO DE 10 ml DE MUESTRA DEL PACIENTE PUEDE SER TRANSFERIDO AL Liqui-PREPTM Preservative R Collection Vial.
Después de la recolección, rotular la muestra recogida y enviarla al laboratorio para su procesamiento y diagnóstico.
El exceso de muestra del paciente debe ser almacenado añadiendo 1 parte de muestras a 2 partes de Liqui-PREPTM Preservative Solution en 2°-8° C.
STEP 2: PRESERVATION



ALLOW A MINIMUM OF 1 HOUR FOR THE SPECIMEN TO BE PRESERVED (Fixation) BEFORE PROCESSING THE SPECIMEN.
After preservation, mix the specimen very well using a vortex mixer (at least 1,000 RPM) for 1 to 2 minutes.
Proceed immediately to Cleaning and Concentration.
PASO 2: PRESERVACION



PERMITA UN MINIMO DE 1 HORA PARA QUE LA MUESTRA SE PRESERVE (fijación) ANTES DE PROCESARLA.
Después de preservación, mezclar la muestra muy bien usando un mezclador vortex (por lo menos 1,000 rpm) durante 1 a 2 minutos.
Proceda inmediatamente a la limpieza y la concentración.
STEP 3 & 4: CLEANING AND CONCENTRATION


Pipette 5mL of Liqui-PREPTM Cleaning Solution R into the 30ml centrifuge tube supplied in the kit.
Pour the entire specimen from the specimen collection vial into the centrifuge tube.



Pipetear 5 ml de Liqui-PREPTM Cleaning Solution R en el tubo de centrífuga de 30mL tapado suministrado en el kit.




Place the 30mL centrifuge tube with the specimen and the cleaning solution into the swinging bucket centrifuge.
Centrifuge for 10 minutes at 1,000 x g force.
After centrifugation, remove the cap and invert the tube keeping it upside down until all the supernatant has poured from the tube.
PASO 5 Y 6: CENTRIFUGACION



Coloque el tubo de centrífuga de 30 mL con la muestra y la solución de limpieza en la centrífuga de cubo oscilante.
Centrifugar durante 10 minutos a 1,000 x g fuerza.

STEP 7 & 8: ENCAPSULATION AND ADHERENCE




Mix the cellular pellet very well for as much as 1 minute to ensure the pellet is homogeneous.
Pipette 100µl of the homogeneous pellet into another clean tube.
To this new dilution tube, add 500µl of Liqui-PREPTM Cellular Base Solution R.
Add 5mL of the standard Liqui-PREPTM Preservative Solution to the 30ml centrifuge tube to preserve the specimen for future testing. Mix, re-cap and store at 2o-8o C.
PASO 7 Y 8: ENCAPSULACION Y ADHESION




Mezclar el botón celular muy bien tanto como 1 minuto para asegurar el botón es homogéneo.
Pipetear 100μl del botón homogénea en otro tubo limpio.
Con este nuevo tubo de dilución, añada 500μl de Liqui-PREPTM Cellular Base Solution R.
Añadir 5ml de Liqui-PREPTM Preservative Solution en el tubo de centrífuga de 30ml a preservar la muestra para futuras pruebas. Mezclar, volver a la tapa y almacene a
2°-8° C.
STEP 9: SLIDE PREPERATION


Using a pipette, take 50µl of the cell pellet/cellular base dilution from the tube and place it on a clean glass microscope slide.
Allow the slide to dry, stain and read the slide.
PASO 9: PREPARACION DE DIAPOSITIVA


Utilizando una pipeta, tomar 50μl del tubo de la dilución de botón celular/base celular y colocar en una diapositiva de vidrio de microscopio limpio.
